Ο Παντελής Παυλάκης σπούδασε στην Ιατρική Σχολή του Εθνικού και Καποδιστριακού Πανεπιστημίου Αθηνών. Μετά την αποφοίτησή του, εκπόνησε διδιακτορική διατριβή στο Εργαστήριο Παθολογικής Φυσιολογίας της ίδιας Σχολής. Ύστερα μετέβη στις ΗΠΑ όπου ειδικεύθηκε πλήρως στην Νευρολογία στο Boston University. Μετά το πέρας της ειδίκευσής του, εξειδικεύτηκε στα Νευρομυϊκά νοσήματα, την Ηλεκτρομυογραφία και την Αυτοάνοση Νευρολογία στο νοσοκομείο Hospital for Special Surgery της Ιατρικής Σχολής του Πανεπιστημίου Cornell. Κατόπιν εξετάσεων, έλαβε πιστοποίηση στη Νευρολογία και τα Νευρομυϊκά Νοσήματα από το American Board of Psychiatry and Neurology. 


Μετά το πέρας της εκπαίδευσής του, παρέμεινε στο ίδιο νοσοκομείο ως Επιμελητής Νευρολόγος και Επίκουρος Καθηγητής Νευρολογίας Ιατρικής Σχολής του Πανεπιστημίου Cornell. Διετέλεσε διευθυντής των ειδικών ιατρείων Πλάγιας Μυατροφικής Σκλήρυνσης και Μυϊκής Δυστροφίας (ALSA & MDA clinics). Συμμετείχε, ως κύριος ερευνητής, σε κλινικές μελέτες νευρομυϊκών νοσημάτων συμπεριλαμβανομένων της πλάγιας μυατροφικής σκλήρυνσης, μυασθένειας και χρόνιας φλεγμονώδους απομυελινωτικής πολυνευροπάθειας.  Επίσης δίδασκε φοιτητές, ειδικευόμενους και εξειδικευόμενους Νευρολογίας της Ιατρικής Σχολής του Πανεπιστημίου Cornell. Έχει συγγράψει επιστημονικά άρθρα σε διεθνή περιοδικά και κεφάλαια σε βιβλία. 


Μετά την επιστροφή του στην Ελλάδα, διατηρεί ιδιωτικό ιατρείο στους Αμπελοκήπους και είναι Συνεργάτης του Ιατρικού Αθηνών, στο Μαρούσι.

Επισκεφθήκαμε τον κ. Παντελή Παυλάκη στο ιατρείο του για τη μητέρα μου, η οποία είναι ηλικιωμένη και αντιμετώπιζε κάποια νευρολογικά προβλήματα. Από την πρώτη στιγμή, η προσέγγισή του ήταν επαγγελματική, αλλά και πολύ ανθρώπινη, δείχνοντας πραγματικό ενδιαφέρον για τις ανάγκες της μητέρας μου. Ο γιατρός αφιέρωσε όσο χρόνο χρειαζόταν για να μας εξηγήσει αναλυτικά τα αποτελέσματα των εξετάσεων και να μας καθησυχάσει με τις σαφείς και τεκμηριωμένες απαντήσεις του. Η φροντίδα και η προσοχή που έδειξε σε αυτή την τόσο ευαίσθητη περίπτωση ήταν πραγματικά σημαντική!
